摘要:
表面等离子共振成像(SPRI)是一种高灵敏度,高通量的,可用于实时分析生物蛋白间的相互作用的新技术。 由于高通量,不需要额外标记等优点,该技术广泛应用于药物筛选、食品检测、环境监测等领域。 实现 SPRI 视频自动化分析的关键是自动定位 SPRI 视频图像中的蛋白阵列,为解决该问题提出了基于重映射的 SPRI 视频自动找点的算法。 算法需要输入生物芯片的打印效果图和 SPRI 视频图像,处理过程分为六个部分:截取,边缘检测,椭圆拟合,阵列识别,坐标映射和自动校准,输出 SPRI 视频中蛋白点阵列的坐标矩阵。 算法基于将打印效果图中的蛋白点坐标重映射为 SPRI 视频中蛋白点坐标思路实现。 实验结果表明,该算法能准确定位 SPRI 视频中的蛋白阵列,不受蛋白点的阵列模式和 SPRI 视频中噪音的影响。 并且算法定位 SPRI 视频中蛋白阵列耗时短,支持在嵌入式设备上实时分析 SPRI 视频.
Abstract:
Surface plasm on resonance imaging (SPRI) is a new technology with high sensitivity and high through put,which can be used to analyze the interaction between proteins in real time. Due to label free and high through put,it is widely used in drug screening,food detection,environmental monitoring and other fields. The key to automatic analysis of SPRI video is to automatically locate the protein array in the SPRI video image. In order to solve this problem,an algorithm based on remapping? ?for automatic spot identification of SPRI video is proposed. The algorithm needs to input the printed image of biochip and frame of SPRI video. The processing can be divided into six parts:image crop,edge detection,ellipse fitting,array recognition,remapping and automatic alignment,and output the coordinate matrix of protein point array in SPRI video. The algorithm is based on the idea of remapping the coordinates of the protein points in the printed renderings to the coordinates of the protein points in the SPRI video. The experiment shows that the proposed algorithm can locate the protein array in the SPRI video accurately,regardless of the pattern of protein points and the noise in the SPRI video. It also takes less time to locate the protein array in SPRI video,and supports real-time analysis of SPRI video on embedded devices.
